ABSTRACT

Neurotropic viruses can cross the otherwise dynamically regulated blood-brain barrier (BBB) and affect the brain cells. Zika virus (ZIKV) is an enveloped neurotropic Flavivirus known to cause severe neurological complications, such as encephalitis and fetal microcephaly. In the present study, we employed human brain microvascular endothelial cells (hBMECs) and astrocytes derived from human progenitors to establish a physiologically relevant BBB model. We used this model to investigate the effects of ZIKV envelope (E) protein on properties of cells comprising the BBB. E protein is the principal viral protein involved in interaction with host cell surface receptors, facilitating the viral entry. Our findings show that the presence of ZIKV E protein leads to activation of both hBMECs and astrocytes. In hBMECs, we observed a decrease in the expression of crucial endothelial junction proteins such as ZO-1, Occludin and VE-Cadherin, which are vital in establishment and maintenance of the BBB. Consequently, the ZIKV E protein induced changes in BBB integrity and permeability. We also found upregulation of genes involved in leukocyte recruitment along with increased proinflammatory chemokines and cytokines upon exposure to E protein. Additionally, the E protein also led to astrogliosis, evident from the elevated expression of GFAP and Vimentin. Both cell types comprising the BBB exhibited inflammatory response upon exposure to E protein which may influence viral access into the central nervous system (CNS) and subsequent infection of other CNS cells. Overall, our study provides valuable insights into the transient changes that occur at the site of BBB upon ZIKV infection.

ABSTRACT

Positive and negative artifacts of particle-phase organic carbon (p-OC) and the polycyclic aromatic hydrocarbons (PAHs) in gasoline direct injection (GDI) engine exhaust particulate matter (PM) were assessed using an integrated organic gas and particle sampler (IOGAPS). Three configurations (denuder + sorbent impregnated filters (SIFs), upstream Zefluor filter + denuder + SIFs, and standard filter pack + SIFs) were used to collect GDI exhaust samples at cold start and highway cruise operating conditions with no aftertreatment. Approximately 35% of the measured GDI p-OC was attributed to positive artifacts; negative artifacts were not detectable due to low overall SVOC concentrations. GDI engine exhaust PAH concentrations were approximately 10 times higher during cold start than highway cruise. At highway cruise, pyrene and fluoranthene were the dominant PAHs in the undenuded filter pack; downstream of the denuder benzo(a)anthracene was the dominant PAH. From a comparison of our findings to published PAH emission factors we estimate that three-way catalyst conversion efficiencies of PAHs were approximately 80% for 3 of the 15 PAHs measured during highway cruise operation. These conversion efficiencies may be considerably lower during cold start operation when the three-way catalyst has not reached its operating temperature. Our previous work showed that adverse biological responses to GDI engine exhaust exposure may be dominated by the particle phase when measured downstream of a Teflon filter. Understanding the partitioning characteristics of PAHs may help elucidate specific PAHs contributing to this effect.

ABSTRACT

Tourism can form an important component of a nation's GDP, and Vietnam is among the most visited countries in Southeast Asia. Most studies on personal exposure focus on the general population, or occupational cohorts with exposure to specific pollutants. However, short-term exposure to air pollutants while visiting regions with high levels of air pollution can lead to acute health effects. A personal exposure study was conducted across three cities in Vietnam to estimate exposure to particulate matter (PM2.5) and black carbon for tourists. Measurements were conducted during the wet season in 2014 in Ho Chi Minh City, Da Lat and Nha Trang using portable instrumentation. Average 24-hr PM2.5 and BC exposures were estimated as 18.9±9.24 and 3.41±1.33µg/m3 and among the three cities, Ho Chi Minh was found to have the highest PM2.5 concentrations. Environmental tobacco smoke, commuting and street food stands were found to contribute to highest levels of exposure to PM2.5 and BC across all cities.

ABSTRACT

Personal exposure (PE) to air pollutants is driven by a combination of pollutant concentrations in indoor and outdoor environments, and time-activity pattern of individuals. The objectives of this study were to estimate personal exposure to PM2.5 and black carbon (BC), and assess the representability of ambient air quality monitoring stations to serve as surrogates for PE in New Delhi. Personal exposure to air pollutants (PM2.5-PE and BCPE) was measured using portable, battery-operated instruments (PM2.5- pDR1500 and BC- microAethalometer AE51) in a small cohort of healthy adults (n=12 in summer, n=6 in winter) with no occupational exposure. Average PM2.5-PE and BCPE (µg/m3) were 53.9±136 and 3.71±4.29 respectively, in summer and 489.2±209.2 and 23.3±14.9 respectively, in winter. Activities associated with highest exposure levels were cooking and indoor cleaning for PM2.5, and commuting for BC. Within transport microenvironments, autorickshaws were found to be the most polluted, and lowest BC exposure was registered in public buses. Comparison of fixed-site ambient monitoring data showed a higher correlation with personal exposure dataset in winter compared to summer (r2 of 0.51 (winter) and 0.21 (summer); 51% (winter) and 20% (summer)). This study highlights the need for detailed assessment of PE to air pollutants in Indian cities, and calls for a denser network of monitoring stations for better exposure assessment.

ABSTRACT

PM2.5 concentrations were measured in residential indoor environment in slums of central India during 2012-2013. In addition, a suite of chemical components including metals (Al, K, Ca, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Cd, Pb, Mo, Se, Sb, Na, Mg, K and Hg), ions (Na+, Mg2+, K+, Ca2+, F-, Cl-, NH4+, NO3- and SO42-) and carbon (OC and EC) were analyzed for all samples. Indoor PM2.5 concentrations were found to be several folds higher than the 24-h national ambient air quality standard (60 µg/m3) for PM2.5 in India, and the concentrations were found to vary from season to season. Mass closure was attempted for PM2.5 data, and close to 100 % mass was accounted for by organic matter, crustal material, secondary organic and inorganic aerosols and elemental carbon. Additionally, carcinogenic and non-carcinogenic health risks associated with exposure to indoor PM2.5 (inhalation, dermal and ingestion) were estimated and while exposures associated with dermal contact and ingestion were found to be within the acceptable limits, risk associated with inhalation exposure was found to be high for children and adults. Elements including Al, Cd, Co, Cr, Mn, Ni, As and Pb were present in high concentrations and contributed to carcinogenic and non-carcinogenic risks for residents' health. Results from this study highlight the need for efforts to reduce air pollution exposure in slum areas.

ABSTRACT

Gasoline direct injection (GDI) engines are increasingly prevalent in the global vehicle fleet. Particulate matter emissions from GDI engines are elevated compared to conventional gasoline engines. The pulmonary effects of these higher particulate emissions are unclear. This study investigated the pulmonary responses induced by GDI engine exhaust using an ex vivo model. The physiochemical properties of GDI engine exhaust were assessed. Precision cut lung slices were prepared using Balb/c mice to evaluate the pulmonary response induced by one-hour exposure to engine-out exhaust from a laboratory GDI engine operated at conditions equivalent to vehicle highway cruise conditions. Lung slices were exposed at an air-liquid interface using an electrostatic aerosol in vitro exposure system. Particulate and gaseous exhaust was fractionated to contrast mRNA production related to polycyclic aromatic hydrocarbon (PAH) metabolism and oxidative stress. Exposure to GDI engine exhaust upregulated genes involved in PAH metabolism, including Cyp1a1 (2.71, SE=0.22), and Cyp1b1 (3.24, SE=0.12) compared to HEPA filtered air (p<0.05). GDI engine exhaust further increased Cyp1b1 expression compared to filtered GDI engine exhaust (i.e., gas fraction only), suggesting this response was associated with the particulate fraction. Exhaust particulate was dominated by high molecular weight PAHs. Hmox1, an oxidative stress marker, exhibited increased expression after exposure to GDI (1.63, SE=0.03) and filtered GDI (1.55, SE=0.04) engine exhaust compared to HEPA filtered air (p<0.05), likely attributable to a combination of the gas and particulate fractions. Exposure to GDI engine exhaust contributes to upregulation of genes related to the metabolism of PAHs and oxidative stress.

ABSTRACT

Organic aerosol composition in the urban atmosphere is highly complex and strongly influenced by vehicular emissions which vary according to the make-up of the vehicle fleet. Normalized test measurements do not necessarily reflect real-world emission profiles and road tunnels are therefore ideal locations to characterise realistic traffic particle emissions with minimal interference from other particle sources and from atmospheric aging processes affecting their composition. In the current study, the composition of fine particles (diameter ≤2.5 µm) at an urban background site (Elms Road Observatory Site) and a road tunnel (Queensway) in Birmingham, UK, were analysed with direct infusion, nano-electrospray ionisation ultrahigh resolution mass spectrometry (UHRMS). The overall particle composition at these two sites is compared with an industrial harbour site in Cork, Ireland, with special emphasis on oxidised mono-aromatics, polycyclic aromatic hydrocarbons (PAHs) and nitro-aromatics. Different classification criteria, such as double bond equivalents, aromaticity index and aromaticity equivalent are used and compared to assess the fraction of aromatic components in the approximately one thousand oxidized organic compounds at the different sampling locations.

ABSTRACT

Air pollution poses a critical threat to human health with ambient and household air pollution identified as key health risks in India. While there are many studies investigating concentration, composition, and health effects of air pollution, investigators are only beginning to focus on estimating or measuring personal exposure. Further, the relevance of exposures studies from the developed countries in developing countries is uncertain. This review summarizes existing research on exposure to particulate matter (PM) in India, identifies gaps and offers recommendations for future research. There are a limited number of studies focused on exposure to PM and/or associated health effects in India, but it is evident that levels of exposure are much higher than those reported in developed countries. Most studies have focused on coarse aerosols, with a few studies on fine aerosols. Additionally, most studies have focused on a handful of cities, and there are many unknowns in terms of ambient levels of PM as well as personal exposure. Given the high mortality burden associated with air pollution exposure in India, a deeper understanding of ambient pollutant levels as well as source strengths is crucial, both in urban and rural areas. Further, the attention needs to expand beyond the handful large cities that have been studied in detail.

ABSTRACT

Most studies of road dust composition have sampled a very wide range of particle sizes, but from the perspective of respiratory exposure to resuspended dusts, it is the PM10 fraction which is of most importance. The PM10 fraction of road dust samples was collected at two sites in Birmingham, UK (major highway and road tunnel) and one site in New Delhi, India. Dust loadings were found to be much higher for New Delhi compared to Birmingham, while concentrations of several species were much higher in the case of Birmingham. Detailed chemical source profiles were prepared for both cities and previously generated empirical factors for source attribution to brake wear, tyre wear, and crustal dust were successfully applied to the UK sites. However, 100% of the mass for the Indian site could not be accounted for using these factors. This study highlights the need for generation of local empirical estimation factors for non-exhaust vehicle emissions. A limited number of bulk road dust and brake pad samples were also characterized. Oxidative potential (OP) was also determined for a limited number of PM10 and bulk road dust samples, and Cu was found to be a factor significantly associated with OP in PM10 and bulk road dust.
